Layton and Luke are caught up in an adventure, when a masked figure steals an entire opera house and forces those in attendance to play a high-stakes game. The winner will receive eternal life, but it could mean death for the losers.

The Nohara family encounters an alien named Shiriri who accidentally turns the parents into kids. They must find Shiriri's father, leading to an adventure spanning Japan.

The film features Shinnosuke as a ninja, and will reveal if Shinnosuke is the real son of Hiroshi and Misae.
